The story Dhama Chaukdi is a movie of four underworld dons. The underworld dons are: Mani Aiyer from Madras (Mukesh Tiwari), Lallan Pyare from UP (Deepak Chauhan), Pappu Yadav from Bihar (Akshay Yadav), and Sanjay Apte from Aamchi Mumbai (Yadhuveer Yadav). These four dons are the enemies in Mumbai. These 4 enemies are the haters of each other. They cannot contact each other neither they cannot bear each other.

The new police officer changes every ABCD of their business. The police officer destroys and bans every illegal work. These dons have a police officer as their common enemy. These four join hands with each other to face that common man. The four try to unit with each other and that is how, the film begins. The film takes many interesting turn and twist to show. The four don soon joins and enters the Bollywood. The story of the movie is further taken to show how they shoot and how their profession is deliberately converted to work for themselves.
